Mechanism

GHK is a tripeptide with the sequence Gly-His-Lys. It occurs naturally in human plasma at concentrations of ~200 ng/mL in young adults, declining to ~80 ng/mL by age 60. The histidine residue gives GHK strong copper(II) binding affinity, and the GHK-Cu complex is the biologically active form.

The copper-binding is the key to the molecule’s pharmacology. GHK-Cu acts as a copper-delivery vehicle to specific cellular contexts where copper is needed for enzymatic activity (lysyl oxidase in collagen cross-linking, superoxide dismutase, etc.). It also has direct receptor effects independent of the copper, but these are less well-characterized.

Documented effects in cell culture and skin biopsy studies:

  • Stimulation of collagen and elastin synthesis in fibroblasts
  • Antioxidant activity through copper-dependent enzymes
  • Modulation of inflammatory cytokine production
  • Promotion of dermal extracellular matrix repair

The mechanism is most relevant in tissues where copper-dependent processes drive function: skin, vasculature, and wound-healing contexts. The mechanism’s relevance to systemic injection is less clear because the route of delivery affects which tissues see meaningful concentrations.


What the evidence shows

Topical use (skin care and dermatology):

  • Multiple published studies showing improvements in fine wrinkles, skin firmness, and overall appearance with topical GHK-Cu cream applied over 12+ weeks
  • Wound healing acceleration in chronic ulcer studies (limited but published)
  • Incorporation into prescription dermatology products (Procyte and successors) and many cosmetic formulations

The topical evidence is real but mostly in small studies (20–100 subjects) without large pivotal trials. It’s enough to justify the cosmetic and dermatology incorporation; not enough for any clinical claim of disease-modifying effect.

Injectable use (systemic):

  • No published RCT evidence for systemic anti-aging, longevity, recovery, or athletic-performance claims
  • Some animal data on tissue regeneration with injection in specific injury models
  • Hair-restoration claims rest on small clinical observation studies, not RCTs

The contrast matters:

A topical cream with characterized local skin effects is a fundamentally different product from an injectable peptide-copper complex distributed systemically. The marketing often blurs this — “GHK-Cu has clinical evidence” is true for the topical use case, but is not true for the injection use case the gray-market sells.


Dosing literature

Topical (cosmetic / dermatology):

GHK-Cu is included in cosmetic formulations at ~0.05–0.5% concentration. Prescription dermatology preparations have been formulated at higher concentrations.

Injectable (gray-market):

No standard or evidence-based protocol. User reports describe:

  • Subcutaneous injection: 1–5 mg per dose
  • Frequency: daily to weekly
  • Cycle structure: highly variable

We do not endorse any injectable dose. The numbers above describe what users report, not a clinical recommendation.


Risks and adverse events

Topical use (well-characterized):

  • Generally well-tolerated
  • Occasional mild contact dermatitis or skin irritation
  • Pigmentation changes uncommon but reported

Injectable use (less characterized):

  • Injection-site reactions (often more severe than other peptides because the copper component can be irritating)
  • Local burning or pain at injection site
  • Skin discoloration at injection site (possibly from copper)

Chronic copper-exposure concerns:

Copper is biologically essential but tightly homeostatically regulated. Chronic supraphysiologic injection of copper-bearing compounds raises concerns about:

  • Wilson’s disease-like accumulation in unusual exposure patterns
  • Hepatic copper deposition with very-long-term high-dose use
  • Disruption of copper/zinc balance (copper supplementation can affect zinc status)

These concerns are theoretical for the typical user pattern (months, not years; low daily doses) but become more relevant with chronic high-dose use over years. The Wilson’s disease connection is mostly relevant in patients with the genetic predisposition; for genetically normal users, the risk is much lower but not zero with chronic high-dose exposure.

Quality concerns:

The copper component adds a quality dimension that pure-peptide products don’t have: GHK-Cu products vary in their actual copper-to-peptide ratio, which determines biological activity. Without analytical verification, you can’t know what you’ve bought.

The cosmetic and topical regulatory status is well-established. The injectable status is everywhere “not approved as a medicine.”


Where to get it

For topical use: through cosmetic skincare retailers or dermatology prescription. The topical use case is the only one we’d recommend, and the access path is through normal skincare channels.

For injectable use: we do not route readers to a fulfillment partner. The combination of unapproved regulatory status, chronic-copper-exposure concerns, and absence of efficacy data for injectable systemic effects puts it outside what we’ll endorse.
